The CRISPR-Cas system is an adaptive immune defense system developed by bacteria and archaea during biological evolution for the removal of exogenous viruses or plasmid nucleic acids. Nucleic Acids Detection by CRIPSR-Cas System

When the target nucleic acid is present in the system to be detected, the Cas protein specifically recognizes the target sequence under the guidance of the guide RNA and forms a complex, which is then stimulated with the "trans-cutting activity" against the cut single-stranded nucleic acid, shredding the fluorescent labeled probe in the system and emitting a fluorescent signal to complete the detection process. The presence or absence of the target nucleic acid can be determined by detecting whether the system is luminescent.

The CRISPR-Cas9 system has the ability to precisely identify and cleave specific DNA sequences, and the combination of this property and isothermal amplification technology has developed some molecular assays that enable highly sensitive nucleic acids detection. The CRISPR-associated protein Cas12, a type V of the class II CRISPR system, is an RNA-directed nucleic acid endonuclease with incidental cleavage activity. It can specifically cleave target nucleic acids under the guidance of crRNA with an accompanying non-specific cleavage activity of ssDNA. Therefore, it can be used for a variety of nucleic acids assays. Cas13 is an RNA-guided nucleic acid endonuclease that specifically cleaves single-stranded target nucleic acids under the guidance of crRNA and can remain active and cleave other non-target nucleic acid after completion of cleavage. This property can be used for the detection of nucleic acids in bioanalysis.
