RNA Detection by CRISPR-Cas System

When the target RNA is present in the system to be tested, the Cas protein specifically recognizes the target sequence under the guidance of the guide RNA and forms a complex, which in turn is stimulated with "trans cleavage activity" against the cleaved single-stranded nucleic acid, shredding the fluorescent labeled probe and emitting a fluorescent signal. Finally, the presence of target RNA is determined by detecting whether the system is luminescent or not.

CRISPR-Cas12 CRISPR-Cas13
The CRISPR-associated protein Cas12 is a type V of the class II CRISPR system, an RNA-guided nucleic acid endonuclease with incidental cleavage activity. It can specifically cleave dsDNA under the guidance of crRNA and is accompanied by a non-specific cleavage activity of ssDNA. After reverse transcription of RNA into DNA, it can be used for RNA detection. Cas13 is an RNA-guided RNA nucleic acid endonuclease that specifically cleaves single-stranded target RNA under the guidance of crRNA and can remain active and cleave other non-target RNA after completion of cleavage. This property can be used for the detection of RNA in bioanalysis.
