DNA Detection Service

The CRISPR-Cas system is an adaptive immune defense system developed by bacteria and archaea during the course of biological evolution. It can be used not only for gene editing, but also shows great potential for DNA detection, which is important in the fields of food safety, environmental microbial contamination detection, and human pathogenic bacteria infection.

DNA Detection by CRISPR-Cas System

The crRNA (CRISPR RNA) is designed to target the target DNA, and then crRNA and Cas protein are added to the detection system. When the target DNA is present, the Cas protein forms a ternary complex with the crRNA and the target DNA, while the complex exercises its trans cleavage activity and cuts the single-stranded DNA labeled with a fluorescent signal (with a luminescent group and a quenching group attached to each end, and the luminescent group can emit light after being cut), thus emitting fluorescence. By detecting the fluorescence, the presence or absence of the target DNA molecule in the system to be tested is known.

The CRISPR-Cas9 system has the ability to precisely identify and cleave specific DNA sequences, and the combination of this property and isothermal amplification technology has developed some molecular assays that enable highly sensitive DNA detection. The CRISPR-associated protein Cas12, a type V of the class II CRISPR system, is an RNA-directed nucleic acid endonuclease with incidental cleavage activity. It can specifically cleave target dsDNA under the guidance of crRNA with an accompanying non-specific cleavage activity of ssDNA. Therefore, it can be used for a variety of DNA assays. Cas13, a type VI CRISPR system-associated protein, is an RNA-guided nucleic acid endonuclease with trans cleavage activity. Based on this property it can be used for DNA detection of multiple sources.
